FAS(TNFRSF6) is a receptor and contains three TNFR-Cys repeats and one death domain. It has been shown that FAS is involved in the physiological regulation of programmed cell death, and has been implicated in the pathogenesis of various malignancies and diseases of the immune system. FADD (adapter molecule) recruits caspase-8 to the activated receptor, the resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases mediating apoptosis. FAS-mediated apoptosis may play a role in the induction of peripheral tolerance, in the antigen-stimulated suicide of mature T-cells, or both.

关于TNFa/TNFSF2：
TNFα is a homotrimer with a subunit molecular mass of 17 kD cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It plays a major role in growth regulation, differentiation, inflammation, viral replication, tumorigenesis, autoimmune diseases and in viral, bacterial, fungal, and parasitic infections. Besides inducing hemorrhagic necrosis of tumors, TNF was found to be involved in tumorigenesis, tumor metastasis, viral replication, septic shock, fever, inflammation, and autoimmune diseases including Crohn’s disease, and rheumatoid arthritis as well as graft-versus-host disease.
