一.细胞描述
Ⅱ型肺泡细胞（ATⅡ）又称颗粒肺泡细胞，散在分布于ATⅠ肺泡细胞（ATⅠ）之间及其相邻的肺泡间隔结合处。其体积较小，呈立方形，表面稍突向肺泡腔。细胞核大而圆，胞质染色较浅淡，胞质中常见空泡。数量较ATⅠ多，ATⅡ占肺泡上皮细胞总数的14%到16%，但仅覆盖5℅的肺泡表面。ATⅡ体积比ATⅠ小很多，人约为900um3 .在细胞游离面有较多的短微绒毛，尤其在细胞边缘部更多。细胞表面有MPA凝集素，对α-半乳糖残基有特异性反应。相邻细胞以紧密连接或中间连接相连，胞质内有较多的线粒体和粗面内质网，还有多泡体、溶酶体和板层体[1]。ATⅡ是肺泡上皮细胞的“干细胞”，它的功能多样：能增殖成新的ATⅡ，还可以分化为其他上细胞如ATⅠ；合成和分泌表面活性物质的功能；肺水转运功能；强大的免疫功能。这些功能与以下疾病有密不可分的关系。
二.细胞特性
(1)细胞来源于人正常肺组织。
(2)细胞鉴定：肺表面活性蛋白A（SP-A）或肺表面活性蛋白C（SP-C）免疫荧光染色为阳性。
(3)经鉴定细胞纯度高于90%。
(4)不含有HIV-1、HBV、HCV、支原体、细菌、酵母和真菌。
(5)细胞生长方式：上皮样，多角形细胞，贴壁培养。
三.细胞运输和保存
视气温状况、运输距离，客户与公司协商后选择下述一种运输方式：
(1)T25培养瓶充满完全培养基后进行常温运输。收到细胞后请镜下观察细胞生长状态，如铺瓶率超过85%请立即进行传代操作，如悬浮的细胞较多，请将培养瓶至于培养箱中静置过夜以帮助未死亡的悬浮细胞能够再次贴壁。
(2)1mL冻存细胞悬液装于1.8mL的冻存管中，置于装满干冰的泡沫保温盒中进行运输。收到细胞后请尽快解冻复苏细胞进行培养，如无法立刻进行复苏操作，冻存细胞可在-80℃的条件下保存1个月。
四.培养建议
人原代II型肺泡上皮细胞的体外培养周期有限。建议使用本公司配套的原代细胞专用培养基和正确的培养方法来解冻、传代，以此保证细胞具备良好的增殖和分化能力。推荐使用：原代上皮细胞培养基（货号:PM-001）。
五.产品用途
