Neuropixels
 
Neuropixels是目前全球流行的基于CMOS芯片的在体多通道记录设备。不同于传统的微丝电极和硅电极,Neuropixels在探针部位就已经完成信号的放大和数模转换,使得采集信号更稳定;能够集成更多记录位点和记录通道。
 
经典的Neropixels 1.0(NP 1.0)探针在10mm长的电极柄上均匀分布了960个记录位点,这一独特的设计非常适合贯穿多个脑区,记录大量脑区的神经元放电。

 

新推出的Neuropixels 2.0(NP 2.0)探针,使Neuropixels的功能进一步升级,每个探针拥有四个电极柄,每根电极柄的记录位点数量增加到1280个,因此每根探针有5120个记录位点,此外,探针和headstage的体积更小重量更轻,更适合在自由活动的动物上进行数据采集。

想要最大程度发挥Neuropixels全脑大尺度神经检测的优势,就不得不提New Scale Technologies公司研发的​高精度多探针操纵器系统(Multiprobe Manipulator, MPM)。
 
2023年,骆利群教授在《Nature》杂志上发表的论文就用到了Neuropixels+MPM的组合。研究团队在一个动物上同时植入四根探针,通过多次植入,实现了对全脑多个脑区的广泛覆盖。通过深入分析各脑区的单细胞放电活动和动物行为间的关系,揭示了大脑编码冲突性选择的内在机制。
 
为什么需要MPM
Neuropixels虽然功能强大,但是利用传统的立体定位仪进行电极的植入很难发挥Neuropixels的优势,并且会带来很多问题。
1、Neuropixels电极比普通电极长很多,并且要求包括电极尖端在内的整根电极柄都能植入至对应的脑区,对定位精度的要求比传统电极高出很多,传统立体定位仪的精度很难达到要求;
2、Neuropixels电极的高效使用方式,通常需要使其一次性穿过准备记录的多个脑区核团,植入的路径通常并不垂直于颅顶面,传统的立体定位仪在进行XZ和YZ两个面均有角度的植入时,需要复杂的计算,实现难度往往很大;
3、为实现更多的通道数,很多实验室都会设计一次植入多根电极,并且要保证这些电极互相不会产生干扰,用传统立体定位仪很难实现;    
4、Neuropixels电极十分昂贵,如何保证其植入的成功率,并减少电极的损伤,是开展实验前需要认真考虑的实际问题。
 
MPM的优势
作为Neuropixels官方推荐的植入平台,MPM解决了Neuropixels探针植入过程中的很多难点,包括:多电极植入、倾斜植入、准确性、稳定性和安全性等。
1、支持多达八根探针的同时植入。
2、独一无二的虚拟植入系统。MPM开发了其最有特色的虚拟植入软件系统,很好地解决了多探针植入轨迹规划的难题。
首先在NTE轨道规划软件中设计探针植入的位置和深度,使得探针可以尽可能多地贯穿感兴趣脑区。
结合NTE软件中获得的参数,在虚拟植入系统中反复调试最佳的植入轨迹,并获得合适的平台参数。这使得实际植入过程变得十分便捷,并可以有效避免探针碰撞引起的损伤。
3、 统一坐标系,前后囟校准。
MPM软件针对每一根探针可进行前后囟校准,并建立统一的坐标系,无需担心动物头部的轻微歪斜,进一步确保倾斜植入的准确性。
4、提供多种移动探针的便捷方法。可同时使用摇杆、手柄和软件控制微操,使用更自由便捷。
5、实时呈现探针植入全过程。植入系统与NTE轨道设计软件联动,可实时看到每根探针接触到的脑区,结合实时电生理信号,帮助判断植入准确性。

Neuropixels is a globally popular CMOS-based in vivo multichannel recording device. Unlike traditional microwire or silicon electrodes, Neuropixels performs signal amplification and analog-to-digital conversion directly on the probe, resulting in more stable signal acquisition and enabling higher-density recording sites and channels.  
 
The classic Neuropixels 1.0 (NP 1.0) probe features 960 recording sites evenly distributed along a 10mm shank, making it ideal for spanning multiple brain regions and capturing neuronal activity across large-scale neural circuits.  
 
The newly released Neuropixels 2.0 (NP 2.0) further enhances functionality, with four shanks per probe and 1,280 recording sites per shank (5,120 total per probe). Additionally, the probe and headstage are smaller and lighter, making them better suited for freely moving animals.

 Why MPM?  
 
While Neuropixels is powerful, traditional stereotaxic systems struggle to maximize its potential and introduce several challenges:  
 
1. Higher precision requirements – Neuropixels probes are much longer than conventional electrodes, requiring the entire shank (including the tip) to reach target regions accurately. Traditional stereotaxic systems often lack the necessary precision.  
2. Complex trajectory planning – Optimal Neuropixels use often requires oblique insertions through multiple brain regions. Traditional systems struggle with dual-plane angle adjustments (XZ & YZ), making implantation difficult.  
3. Multi-probe coordination – Many labs implant multiple probes simultaneously to increase channel count, but avoiding probe collisions is nearly impossible with traditional systems.  
4. Probe safety – Neuropixels probes are expensive. Ensuring high implantation success rates and minimizing damage are critical practical concerns.
