人Ⅳ型胶原介绍:
IV型胶原蛋白是在分离的多种上皮和内皮细胞的细胞外基膜中发现的主要胶原。它存在于真皮与表皮交界的地方,是致密板的一个重要组成部分。它含有两个α1状和一种α2样链的异源分子。IV型胶原蛋白是从人胎盘中分离并提纯的。每小瓶该产品为含5mg无菌的IV型胶原冻干粉末。IV型胶原蛋白通常作为薄涂层的组织培养表面。该产品通常用于在体外作为衬底支架,以增强细胞附着,粘附和增殖。IV型胶原可用于培养上皮细胞,内皮细胞,肌肉,神经和许多其他类型的细胞。此外,该产品适合用作底物的胶原酶测定和阳性对照。
Type IV Collagen is the primary collagen found in the extracellular basement membranes separating a variety of epithelial and endothelial cells. It is a major component of the dermal-epidermal junction where it is mostly found in the lamina densa. It is a heterotrimeric molecule containing two α1-like and one α 2-like chains.
This Type IV Collagen is isolated from human placenta and is purified using a multi-step process. The product is supplied as a sterile, lyophilized powder containing 5 mg of Type IV collagen per vial. A Certificate of Analysis is available with the purchase of each product.
Type IV Collagen is typically used as a thin coating on tissue culture surfaces. Specific instructions are found in the Directions for Use. This product is generally used in vitro as a substrate scaffold to enhance cell attachment, adherence and proliferation. Type IV collagen may be used to culture epithelial, endothelial, muscle, nerve and many other cell types. Additionally, this product is suitable for use as a substrate for collagenase assays and positive controls.

细胞粘附分析:为了验证细胞附着情况,分别将细胞接种在涂有基质的产品和无基质的表面(阴性对照)。接种细胞的培养基为无血清的DMEM培养基。所有表面均用含1%BSA的溶液封闭。接种细胞在37℃培养1小时,用DPBS清洗。结果表明涂有基质的细胞粘附性显著提高。
牛Ⅰ 型胶原介绍
I型胶原是皮肤、骨、肌腱以及其它纤维结缔组织的重要结构成分,并且它与通过低赖氨酸的羟基化和低碳水化合物的组合物不同。虽然有些种类的胶原蛋白已经确定都包含三螺旋构象的三种多肽链的分子,但在氨基酸序列上存在细微差别。一级结构的氨基酸序列作为主要的主要的重复基序,每三个氨基酸中常以甘氨酸、脯氨酸或是甘氨酸残基前4-羟基脯氨酸形式存在。I型胶原蛋白是两个α1链和一个α2链组成的异源三聚体,在中性pH值和37℃的条件下自发形成了三螺旋支架。
在多细胞生物体内,对细胞生长、分化以及凋亡的控制常依赖于细胞外基质(ECM)的粘附性。由于I型胶原蛋白是细胞外基质的一种丰富的组分,模拟体内细胞环境发现细胞在三维(3D)胶原凝胶中培养比传统的二维系统更好。这已经在心脏和角膜成纤维细胞、肝星状细胞(HSC)和成神经瘤细胞中得到证实。
有些疾病可直接影响细胞外基质的粘附性,而其他的疾病状态可通过改变在ECM中的刚性密度而引起粘附性的改变。由于I型胶原蛋白是细胞外基质粘附性的关键因素,因此它能够在三维胶原凝胶中对研究起到有用的作用。
结构和功能多样化的细胞表面受体通过识别胶原的三级螺旋引起的胶原蛋白亚型存在差异。最知名的胶原受体是整合素α1β1和α2β1。α1β1主要整合平滑肌细胞,而α2β1是上皮细胞和血小板的主要形式。这两种形式在成纤维细胞、内皮细胞、成骨细胞、软骨细胞和淋巴细胞等多种细胞类型中可以表达。某些类型的细胞也可表达其它的胶原蛋白受体,例如糖蛋白VI(GPVI),在血小板中其可以介导粘附和信号传导。其它的胶原蛋白受体包括盘状结构域受体、白细胞相关免疫球蛋白样受体-1和甘露糖受体的家族成员。

Type I Collagen may be used to coat glass cover slips of a variety of cell lines:I型胶原可用于涂层的玻璃盖的多种细胞系
1.稀胶原蛋白(3毫克/毫升提供)溶液在1:10—1:50 13毫米的HCl和涂层表面的无菌盖玻片。 Dilute collagen (supplied at 3 mg/ml) solution at 1:10 - 1:50 with 13 mM HCl and coat surface of sterile glass cover slip.
2. 空气干燥在组织培养罩。Air dry in a tissue culture hood.
3.细胞可直接接种于I型胶原表面。 Cells can be seeded directly on the Type I collagen surface.
注:Ⅰ型胶原涂层的制备使用这种方法可以从长期培养中玻璃表面脱落。Note: Type I Collagen coating preparation using this method may detach from the glass surface in long-term cultures.
I型;Ⅳ型胶原可应用于玻璃盖片的长期培养的第一涂层的聚-L-赖氨酸或多聚鸟氨酸玻璃。这提供了一个更稳定的胶原涂层。Type I & IV Collagen can be applied to glass cover slips for long-term cultures by first coating the glass with Poly-L-Lysine or Poly-L-Ornithine. This provides a more stable collagen coating.
1. 制备多聚赖氨酸或多聚鸟氨酸在0.1-1毫克/毫升。Prepare Poly-L-Lysine or Poly-L-Ornithine at 0.1-1 mg/ml.
2. 添加足够的液池表面无菌盖玻片。Add sufficient solution to pool over surface of sterile glass cover slip.
3. 室温孵育2~24小时。Incubate 2 to 24 hours at room temperature.
4. 穿刺液洗3次,无菌水盖片。Aspirate solution and wash cover slips 3 times with sterile water.
5. 池的胶原蛋白溶液,100克/毫升在表面μ滑盖。Pool collagen solution, 100 μg/ml over surface of cover slip.
6. 孵化4~16小时。Incubate 4 to16 hours.
7. 冲洗一次媒体与种子细胞。inse once with media and seed with cells.
